Newcastle's heavy defeat at Arsenal on Saturday stalled their push for the Premier League's top four, but their focus switches to a place in the last four as they return to North London to face Spurs.
A place in the semi-finals would represent Newcastle's best run in the League Cup since they were runners-up to Manchester City in 1976.
With a Tyne-Wear derby against Sunderland to come on Sunday, this is a pivotal week for the Mapgies.
Manager Alan Pardew said: "We have a really big week in front of us in terms of two great games and we are going to try to win them both. I don't see one of the games as any less important that the other.
"If we win this, we have got all the momentum we need going into the derby. It's a difficult game, but our last two in the cup at Crystal Palace and Manchester City were difficult too, both away games."
Newcastle can take confidence from the fact they have won on their last two visits to White Hart Lane, including a come-from-behind Premier League win in October.
However, four-time League Cup champions Spurs have improved in recent weeks, with 10 points out of a possible 15 in the league, and are seemingly adjusting to the demands of their heavy schedule, with eight Europa League games already played.
Pardew added: "I do think Spurs and us are two of the fittest sides in the division, so if it goes to extra-time - which is a possibility - I think we are both armed for that."
The two teams have already faced each other at the Lane this campaign, a Premier League fixture that the visitors won 2-1 back in October, and Alan Pardew and Co will be confident of repeating that result in north London on Wednesday as the try and stay on track for a long-awaited trophy.

Meanwhile the home team, despite their inconsistent league form of late, will also be eyeing up a possible trophy this season under new head coach Mauricio Pochettino, with Tottenham boosted by another late show in the Premier League at Swansea City on Sunday.
The north Londoners have been making something of a habit of scoring last-gasp winners under the Argentine, although Pochettino will be hoping that his side can get the job done far earlier than normal this time around against the Magpies.
And if they can, Spurs will be one step closer to a fifth League Cup crown.
